Astroglial connexins in epileptogenesis

چکیده
The astroglial network connected through gap junctions assembling from connexins physiologically balances the concentrations of ions and neurotransmitters around neurons. Astrocytic dysfunction has been associated with many neurological disorders including epilepsy. Dissociated result in increased activity connexin hemichannels which triggers brain pathophysiological changes. Previous studies patients animal models epilepsy indicate that reduced junction coupling assembled astrocytes may play an important role epileptogenesis. This abnormal cell-to-cell communication is now emerging as feature pathologies being considered a novel therapeutic target for controlling In particular, candidate drugs ability inhibition hemichannel enhancement formation should be explored to prevent epileptogenesis control

New Insights on Astroglial Connexins as a Therapeutic Target for Alzheimer's Disease
Alzheimer’s disease (AD) is the leading cause of dementia in the elderly and characterized by progressive memory loss, behavioral deficits and significant personality alterations [1,2]. The histopathological hallmarks of AD include Aβ plaques, neurofibrillary tangles, neuronal death and synapse loss. One significant feature of Aβ accumulation is the strong reactive gliosis associated with the A...
متن کاملHypothalamic astroglial connexins are required for brain glucose sensing-induced insulin secretion.
Hypothalamic glucose detection participates in maintaining glycemic balance, food intake, and thermogenesis. Although hypothalamic neurons are the executive cells involved in these responses, there is increasing evidence that astrocytes participate in glucose sensing (GS); however, it is unknown whether astroglial networking is required for glucose sensitivity. Astroglial connexins 30 and 43 (C...
متن کاملConnexins in the Nervous System
This chapter reviews the localizations and physiological roles of connexins in neurons and glia of the central and peripheral nervous systems. Cx32 forms gap junctions in noncompact myelin in Schwann cells, which are thought to form a reflexive communication pathway connecting the outer and inner myelin layers. Cx29 is also expressed in myelinating Schwann cells, but does not appear to form gap...
متن کاملImmune mechanisms in epileptogenesis
Epilepsy is a chronic brain disorder that affects 1% of the human population worldwide. Immune responses are implicated in seizure induction and the development of epilepsy. Pre-clinical and clinical evidence have accumulated to suggest a positive feedback cycle between brain inflammation and epileptogenesis. Prolonged or recurrent seizures and brain injuries lead to upregulation of proinflamma...
متن کاملConnexins in leukocytes: shuttling messages?
Gap junctions, formed by the connexin (Cx) protein family, are intercellular channels that permit the cytoplasmic exchange of ions and small metabolites between neighboring cells, a process called gap junction intercellular communication (GJIC). These channels possess unique properties, including distinctive permeabilities for various signaling molecules, which depend on the connexin member(s) ...
